Do accessible rooms have to be reserved exclusively for people with disabilities?
No. It is not discrimination to decline to take a booking from a person requiring an accessible room if all rooms are already booked. It would, however, be prudent management to keep accessible rooms until last so that a person requiring one who seeks a booking can still make one while any rooms remain.
